Recommended Uses: Amplification of the MDM2 gene may be used to distinguish atypical lipomatous sarcoma/well-differentiated liposarcoma (ALT/WDLPS) from benign lipoma. Amplification of the MDM2 gene may also be used to distinguish dedifferentiated liposarcoma (DDLPS) from the subset of pleomorphic lipoma and liposarcoma and myxoid liposarcoma that may share a similar morphology. May be performed in conjunction with other FISH assays or as an independent test.

Specimen Requirements: Paraffin embedded tissue: Five 4-micron sections of tissue containing tumor on immunohistochemistry slides, the middle section stained with H&E.
Specimen Handling & Transport: Store and transport specimens at room temperature.
Collection Kit/Container:
Collection Instructions: FFPE tissue: Tissue specimen should be fixed in 10% neutral buffered formalin for 6-72 hours.
Unacceptable Conditions: FFPE tissue specimen must not be de-calcified.

Required Information: Laboratory regulations require the following minimum information to be provided on the requisition form for a specimen to be accepted for testing: Patient name or unique identifier; date and time of collection, patient date of birth and sex, specimen type/site of collection, test request(s), reason for referral, clinician name and UPIN/NPI, and address for reporting results. Please be certain that name/identifier on the form matches that on the specimen label.
Results Include: Ratio of MDM2 to chromosome 12 centromere signals. MDM2 gene not amplified/MDM2 gene amplified.
Limitations: Negative results do not exclude the diagnosis of well-differentiated liposarcoma/atypical lipomatous tumor. Amplification varies in individual tumors and among different cells in the same tumor.

Methodology: Fluorescence in situ hybridization (FISH) on interphase nuclei (nuc ish) is performed with a DNA probe mix (LSP MDM2/CCP12 FISH Probe kit; CytoTest, Inc.) covering the entire MDM2 gene located at chromosome 12q15 with centromere 12 as the control probe to detect amplification of MDM2.

Amplification of MDM2 is defined as a ratio of MDM2/12CEN >=2.0 and there is an average of >=6.0 MDM2 signals per cell.
Includes: FISH analysis of 60 cells for copy number of the MDM2 gene compared to copy number of chromosome 12 centromere
