Specimen Requirements: Paraffin embedded tissue (FFPE): Seven 4-micron sections on immunohistochemistry slides, the middle section stained with H&E
Specimen Handling & Transport: Store and transport specimens at room temperature (may transport with coolant during hot weather, >85 degrees F). DO NOT FREEZE. The laboratory must receive specimens within 24 to 48 hours of collection.
Collection Kit/Container:
Collection Instructions: FFPE tissue: Tissue specimen should be fixed in 10% neutral buffered formalin for 6-48 hours.
Unacceptable Conditions: Necrotic tissue may not contain nuclei with intact DNA.

Required Information: Laboratory regulations require the following minimum information to be provided on the requisition form for a specimen to be accepted for testing: Patient name or unique identifier; date and time of collection, patient date of birth and sex, specimen type/site of collection, gestational age, test request(s), reason for referral, clinician name and UPIN/NPI, and address for reporting results. Please be certain that name/identifier on the form matches that on the specimen label.
Results Include: Evidence/no evidence of trisomy 13, 16, 18, 21 and/or 22. Sex chromosome signal pattern.
Limitations: FISH results reflect copy number only for the loci of the probes used, and do not rule out isochromosomes or structural abnormalities of chromosomes 13, 16, 18, 21, 22, X, and Y.

Methodology: Fluorescence in situ hybridization (FISH) analysis on interphase nuclei (nuc ish) is performed on paraffin embedded placenta cells using a DNA probe set (AneuVysion: CEP 18/CEP X/CEP Y, LSI 13/21; CEP 16, LSI BCR, Abbott molecular, Inc).
Includes: FISH analysis of paraffin embedded placenta or fetal tissue to determine copy number of chromosomes 13, 16, 18, 21, 22, X and Y.
